A Brief Story About the Development of Membrane Protein Stable Cell Lines
To develop the membrane protein-expressing stable cell line, the encoding gene fragments are cloned into lentiviral vectors with antibiotic resistance genes. The generated recombinant vectors are introduced into mammalian cells, in which the lentivirus particles packaged are collected to infect mammalian cell line system. After enlarging cultivation, the harvested cells are treated with limiting dilution for the screening of stable cells. The infected cells are cultivated in medium supplemented with antibiotics. The cells tested as correct are collected and packaged to stable cell line products, which will be utilized for scaling-up production of the membrane proteins, and the membrane protein dissolving in the cell lysates can be purified using affinity chromatography for the following reconstitution in detergents, liposomes, and nanodiscs to recover the correct folding and the biological activity.
