Isolation of Mouse Bone Marrow-Derived Mesenchymal Stem Cells
Bone marrow-derived mesenchymal stem cells (BMMSCs) are adult stem cells that can differentiate into bone, cartilage, muscle tissue, fat, nerve, and other tissues. Mesenchymal stem cells are characterized by their wide range of sources, convenient acquisition, rapid proliferation, and autologous transplantation, so they are considered ideal seed cells in tissue engineering. However, some studies have shown that the actual content of mesenchymal stem cells in tissue is very low, so how to isolate and obtain high-purity cells and effectively proliferate them is very important. With the continuous improvement in scientific research and technology, researchers have proposed different methods to isolate mesenchymal stem cells. At present, the commonly used methods are differential adhesion, enzyme digestion, density gradient centrifugation, flow cytometry, and magnetic-activated cell sorting.
This article briefly introduces the above methods and summarizes their advantages and disadvantages for your reference.
